In this condensed ~180 second video we are using a Sucrose Density Gradient of different layers (17, 30 and 50%). On top of this gradient a blended pepper emulsion was layered on the surface. The tube is than placed inside the centrifuge for a total of 15 minutes. By speeding up the process we can a quick grasp on how the cells of the pepper move and get concentrated in the different layers and create “banding“ in the Sucrose Density Gradient under 2500 g / RCF.
What hypergravity experiment or separation would you like to see happening inside a centrifuge.
--------- Work in progress ---------
These video’s and project are a work in progress, there are still many adjustments that need to be made to the centrifuge to create an even more optimised recording environment. In regards to the channel format I’m looking into what would be the best way to present these videos, talking and informing about what is happening to the sample from all different (science) perspectives to be able to give the audience a better understanding. Next to that
I’m also looking for help from the (science) community for the following:
- Supporting the project with test samples and / or suggestions on what to centrifuge and/or ways to do this accordingly to simulate existing lab experiments.
- Recommendations, tips and or tricks on how to get better visual results.
- Recommendations, tips and or tricks on how to analyse the samples.
- Leads on a larger and higher RCF centrifuge to expand the capabilities of the project.
If you like to help this project out? I would be more than pleased to hear from you.
00:00 Intro
00:03 Centrifuge ramping up towards 2500g / RCF @4000RPM.
00:05 The cells of the pepper start to diffuse and migrate through the 17% layer.
00:10 The smaller cells are stopped on top of the 30% layer and form a band.
00:15 The larger cells continue to migrate through the 30% layer.
00:20 We can now clearly see 2 bands forming one on top of the 30% & 50% layer
00:30 The first pepper cells start to migrate through the 50% layer.
00:30 The bands start to defuse .
00:40 Cell’s are still migrating through the 30%
01:10 All bands are formed and continue to become more clear and crisp on the edges.
01:25 In the meanwhile we can see the Coriolis force (rotating) clearly being visible in the top layer.
02:30 We can now clearly see several bands are formed at the top of the 17%, 30% and 50%.
02:50 The Centrifuge Continues to spin at 2500G / RCF @4000RPM.
