Antibodies in VR: Designing a Potential Vaccine against Alzheimer’s Disease
In this video, we explore the structures of amyloid-beta (Aβ) protein, specifically the N-truncated species which are implicated in the formation of soluble aggregates and plaques. These plaques are a defining characteristic of Alzheimer’s disease (AD).
Based on structural analysis and structure based design, researchers have discovered a unique conformation and epitope in the N-terminal region of Aβ, which opens up new possibilities for both active and passive immunization strategies against AD. The ’TAPAS’ vaccine is an innovative approach that involves creating a stabilized cyclic form of Aβ1-14, drawing inspiration from its interaction with the murine derived antibody TAP01.
Promising results from experiments involving the TAPAS vaccine in mouse models of AD show notable reductions in neuronal loss, improved brain glucose metabolism, and enhancements in memory function. These findings hold great potential for advancing Alzheimer’s research and potential treatments.
